Abstract Candidemia caused by Candida spp. is a serious threat in hospital settings being major cause of acquired infection and death possible contributor to Covid-19 mortality. incidence has been rising worldwide following increases fungicide-resistant pathogens highlighting the need for more effective antifungal agents with novel modes action. The membrane-bound enzyme alternative oxidase (AOX) promotes fungicide resistance absent humans making it desirable therapeutic target. However, lipophilic nature AOX substrate (ubiquinol-10) hindered its kinetic characterisation physiologically-relevant conditions. Here, we present purification expression recombinant AOXs from C. albicans auris self-assembled proteoliposome (PL) system. Kinetic parameters (K m V max ) respect ubiquinol-10 have determined.
